The 27-year-old, who rushed for 1,287 yards and 12 touchdowns in 15 games last season, has missed only one game the last two seasons after playing only two in 2011 due to a torn ACL.
Charles, a three-time Pro Bowler, is entering his seventh season with the Chiefs. Knile Davis will start in his place.
